چکیده

The g-aminobutyric acid (GABA) is a product of decarboxylation of the amino acid glutamate mediated by the synthesizing enzyme glutamic acid decarboxylase (GAD) (1–3). Although GABA is a major inhibitory neurotransmitter of the brain, it is produced at high levels in pancreatic islets (4). b-Cells store GABA in synaptic-like microvesicles, and upon its secretion, GABA exerts many paracrine functions in pancreatic islets (4). While the total function of GABA in b-cells is incompletely understood (4), its synthesizing enzyme GAD is possibly one of the most significant pancreatic islet b-cell autoantigens (5). GAD is a primary target of autoantibodies, and anti-GAD antibodies are associated with the development of type 1 diabetes (T1D) (5).

In vitro Transdifferentiation of Bone Marrow Stromal Cells into GABAergic-Like Neurons

Background: Cell therapy of many neurodegenerative diseases using bone marrow stromal cells (BMSC) requires the differentiation of BMSC into neuronal subtype. However, the transdifferentiation of BMSC into GABAergic phenotype requires more investigation. Methods: In this study, BMSC of adult female rats were pre-induced into neuroblast-like cells using 1 mM β-mercaptoethanol (βME) and 10 M re...

A New Multistep Induction Protocol for the Transdifferentiation of Bone marrow Stromal Stem Cells into GABAergic Neuron-Like Cells

Background: Bone marrow stromal stem cells (BMSC) are appropriate source of multipotent stem cells that are ideally suited for use in various cell-based therapies. It can be differentiated into neuronal-like cells under appropriate conditions. This study examined the effectiveness of co-stimulation of creatine and retinoic acid in increasing the differentiation of BMSC into GABAergic neuron-lik...

LAT-derived microRNAs in HSV-1 target SMAD3 and SMAD4 in TGF-β/Smad signaling pathway

Background: During its latent infection, HSV-1 produces only a miRNA precursor called LAT, which encodes six distinct miRNAs. Recent studies have suggested that some of these miRNAs could target cellular mRNAs. One of the key cell signaling pathways that can be affected by HSV-1 is the TGF-β/Smad pathway. Herein, we investigated the potential role of the LAT as well as three LAT-derived miRNAs ...

Concise review: pancreas regeneration: recent advances and perspectives.

The replacement of functional pancreatic β-cells is seen as an attractive potential therapy for diabetes, because diabetes results from an inadequate β-cell mass. Inducing replication of the remaining β-cells and new islet formation from progenitors within the pancreas (neogenesis) are the most direct ways to increase the β-cell mass. Stimulation of both replication and neogenesis have been rep...

Potential Pathways to Restore β-Cell Mass: Pluripotent Stem Cells, Reprogramming, and Endogenous Regeneration

Currently available β-cell replacement therapies for patients with diabetes, including islet and pancreas transplantation, are largely successful in restoring normal glucose metabolism, but the scarcity of organ donors restricts their more widespread use. To solve this supply problem, several different strategies for achieving β-cell mass restoration are being pursued. These include the generat...
